Definition
A two-step differential stain using Nuclear Fast Red (Kernechtrot) to stain the sperm head red/pink and Picroindigocarmine to stain the midpiece and tail blue-green. The red-green contrast makes spermatozoa immediately recognisable at 400x.

Common questions
Why use two dyes instead of one?+
A single stain leaves sperm heads and tails the same colour as background debris, so mixed contrast makes each part stand out and lets an examiner distinguish sperm from epithelial cells and other particulate matter quickly.
Does a negative result rule out semen?+
No. Vasectomised men, azoospermic men and degraded samples can leave no intact sperm cells to stain even though seminal fluid is present, so a negative slide is followed up with a biochemical or immunological test for seminal markers.
Can the stain distinguish species?+
No, the dyes bind to the general morphology of any mammalian sperm cell, so a positive stain confirms sperm are present but a separate serological or DNA test is needed to confirm human origin.
