Haem Iron

Definition

The iron atom at the centre of the porphyrin ring in haemoglobin. It catalyses the luminol reaction by cycling between oxidation states during the decomposition of hydrogen peroxide, generating the excited intermediate that emits light.

The light-producing reaction is a catalytic redox process, and it is the iron atom's ability to shuttle between oxidation states that drives the decomposition of hydrogen peroxide into the reactive species needed to excite the luminol molecule, not any structural feature of the surrounding globin protein.

Can substances other than blood trigger luminol's light emission through the same iron pathway?+

Yes. Certain metal ions and bleach-based compounds can catalyse a similar reaction and produce false positives, which is why a luminol reaction is treated as presumptive only and followed by species- and blood-specific confirmatory testing before it is reported as blood.
